Detection of vitamin D2 and D3 in rice flour
Vitamin D (vitamin D) is a steroid derivative with anti-rickets effect, also called anti-rickets vitamin. The most important members of the vitamin D family are D2 and D3. Although some of the vitamin D can be supplemented in the daily diet, a considerable number of people still face the problem of insufficient intake of vitamin D, and various foods for the purpose of adding vitamin D came into being. Diamonsil C18 (2) liquid chromatography column successfully achieved the separation and determination of vitamin D2 and D3 in rice flour, with accurate analysis results. Reproducible characteristics.
Sample preparation
(1) Take 1 g of sample, add 10 mL of water, and mix thoroughly;
(2) Add 1 mL of dimethyl sulfoxide, 5 mL of methanol and 20 mL of n-hexane: methyl tert-butyl ether = 1: 2, vortex to mix for 2 min, centrifuge at 6000 rpm for 2 min, and collect the supernatant;
(3) Add 1 mL of dimethyl sulfoxide, 5 mL of methanol and 10 mL of n-hexane: methyl tert-butyl ether = 1: 2 to the lower residue in sequence, repeat the extraction twice, and combine the three extraction supernatants;
(4) The supernatant is evaporated under reduced pressure at 40 ℃ to near dryness, to be purified.
